Explant innoculation - after decontamination, laminae of unemerged
spear leaves from selected, elite ortet palms are cut into approximately 1 cm x 1 cm fragments
(explants), innoculated onto culture media containing auxin and incubated in the dark.
Callogenesis - calli induced during incubation may be observed about four to six months
after innoculation. Calli are re-cultured onto media containing auxins. |
Embryogenesis - embryoids are formed from calli after about 4 months and
continue till about 12 months. These primary embryoids are transferred to embryoid proliferation
media for multiplication. |
Embryoid proliferation - embryoids proliferate and germinate into
shoots. |
Shoot development - shoots allowed to grow and develop before
root induction. |
Rooted plants - plants with sturdy roots are sent to the nursery to
be weaned and hardened. |
